Technical notes on methylated cucurbiturils

Methylation introduces methyl groups on the glycoluril nitrogen or carbon atoms, weakening intermolecular hydrogen-bond packing and greatly improving water solubility. Symmetrical tetramethyl CB[6] (TMeCB[6]), permethylated CB[5] (Me10CB[5]) and permethylated CB[6] (Me12CB[6]) all allow concentrated aqueous stocks while retaining the rigid cavity and both carbonyl portals, so recognition of cationic and neutral aromatic guests remains comparable to the parent homologues. They suit biological, sensing and materials work in pure water or buffer where an organic co-solvent is undesirable.

Storage, Solubility and Handling Notes

  • Store sealed, dry and protected from light at room temperature; for long-term storage keep at 2–8 °C under dry conditions.
  • Cucurbituril powders are somewhat hygroscopic — reseal promptly after use and record the batch number and opening date.
  • Solubility tips: CB[7] and the modified series dissolve directly in water; for CB[6], dissolve first in formic acid or DMSO, then dilute into the target solvent system.
