Technical notes on methylated cucurbiturils

Methylation adds methyl groups to the glycoluril nitrogen or carbon atoms, disrupting intermolecular hydrogen-bond packing and significantly enhancing water solubility. Symmetrical tetramethyl CB[6] (TMeCB[6]), permethylated CB[5] (Me10CB[5]), and permethylated CB[6] (Me12CB[6]) all support concentrated aqueous stock solutions while preserving the rigid cavity and carbonyl portals. Consequently, their ability to recognize cationic and neutral aromatic guests remains comparable to that of the parent homologues. These derivatives are ideal for biological, sensing, and materials applications in pure water or buffer where organic co-solvents are undesirable.

Storage, Solubility, and Handling Guidelines

  • Store sealed, dry, and protected from light at room temperature. For long-term storage, keep at 2–8 °C under dry conditions.
  • Cucurbituril powders are slightly hygroscopic. Reseal immediately after use and record the batch number and opening date.
  • Solubility tips: CB[7] and the modified series dissolve directly in water. For CB[6], first dissolve in formic acid or DMSO, then dilute into the target solvent system.
  • Filter stock host solutions and use them fresh; for binding experiments, record buffer composition, ionic strength, and pH.
  • These materials are for research use only and must not be used in humans, food, pharmaceutical manufacturing, or clinical diagnostics.
  • Batch-specific purity, water content, and spectral data are available upon request prior to ordering.
